Veröffentlichung Detection and characterization of hepatitis E virus genotype 3 in wastewater and urban surface waters in Germany(2020) Beyer, Sophia; Gnirrs, Regina; Selinka, Hans-Christoph; Szewzyk, RegineIn highly populated areas, environmental surveillance of wastewater and surface waters is a key factor to control the circulation of viruses and risks for public health. Hepatitis E virus (HEV) genotype 3 is considered as an emerging pathogen in industrialized countries. Therefore, this study was carried out to determine the prevalence of HEV in environmental waters in urban and suburban regions in Germany. HEV was monitored in water samples using quantitative RT-PCR (RT-qPCR) and nested RT-PCR without or with virus concentration via polyethylene glycol precipitation or ultracentrifugation. By RT-qPCR, 84-100% of influent samples of wastewater treatment plants were positive for HEV RNA. Genotypes HEV-3c and 3f were identified in wastewater, with HEV-3c being the most prevalent genotype. These data correlate with subtypes identified earlier in patients from the same area. Comparison of wastewater influent and effluent samples revealed a reduction of HEV RNA of about 1 log10 during passage through wastewater treatment plants. In addition, combined sewer overflows (CSOs) after heavy rainfalls were shown to release HEV RNA into surface waters. About 75% of urban river samples taken during these CSO events were positive for HEV RNA by RT-qPCR. In contrast, under normal weather conditions, only around 30% of river samples and 15% of samples from a bathing water located at an urban river were positive for HEV. Representative sample volumes of 1 m3 were taken by applying a fractionated filtration method (500, 100, and 50 (micro)m mesh sizes). The detection of MP mass fractions by thermal extraction desorption-gas chromatography/mass spectrometry (TED-GC/MS) was achieved without the previously required additional sample pretreatment for the first time. Different types of quantification methods for the evaluation of TED-GC/MS data were tested, and their accuracy and feasibility have been proven for real samples. Polyethylene, polystyrene, and polypropylene were identified in effluent samples. The polymer mass content varied significantly between 5 and 50 mg m-3. A correlation between the MP load and the quantity of suspended matter in the WWTP effluents, particle size distribution, particle type, and operation day (i.e., weekday, season, and capacity) was not found. It can be concluded that a meaningful assessment of WWTPs requires a comprehensive sampling campaign with varying operation conditions. © 2022 The AuthorsVeröffentlichung Filling the knowledge gap: A suspect screening study for 1310 potentially persistent and mobile chemicals with SFC- and HILIC-HRMS in two German river systems(2021) Neuwald, Isabelle; Kuckelkorn, Jochen; Muschket, Matthias; Zahn, Daniel; Strobel, ClaudiaPersistent and mobile chemicals (PM chemicals) were searched for in surface waters by hydrophilic interaction liquid chromatography (HILIC) and supercritical fluid chromatography (SFC), both coupled to high resolution mass spectrometry (HRMS). A suspect screening was performed using a newly compiled list of 1310 potential PM chemicals to the data of 11 surface water samples from two river systems. In total, 64 compounds were identified by this approach. The overlap between HILIC- and SFC-HRMS was limited (31 compounds), confirming the complementarity of the two methods used. The identified PM candidates are characterized by a high polarity (median logD -0.4 at pH 7.5), a low molecular weight (median 187 g/mol), are mostly ionic (54 compounds) and contain a large number of heteroatoms (one per four carbons on average). Among the most frequently detected novel or yet scarcely investigated water contaminants were cyanoguanidine (11/11 samples), adamantan-1-amine (10/11), trifluoromethanesulfonate (9/11), 2-acrylamido-2-methylpropanesulfonate (10/11), and the inorganic anions hexafluorophosphate (11/11) and tetrafluoroborate (10/11). 31% of the identified suspects are mainly used in ionic liquids, a chemically diverse group of industrial chemicals with numerous applications that is so far rarely studied for their occurrence in the environment. Prioritization of the findings of PM candidates is hampered by the apparent lack of toxicity data. Hence, precautionary principles and minimization approaches should be applied for the risk assessment and risk management of these substances. The large share of novel water contaminants among these findings of the suspect screening indicates that the universe of PM chemicals present in the environment has so far only scarcely been explored. Dedicated analytical methods and screening lists appear essential to close the analytical gap for PM compounds. © 2021 Published by Elsevier Ltd.Veröffentlichung Molecular detection of cosaviruses in a patient with acute flaccid paralysis and in sewage samples in Germany(2021) Schneider, Julia; Engler, Matthias; Hofmann, Jörg; Selinka, Hans-ChristophCosaviruses (CoSV) were first identified in stool samples collected from non-polio acute flaccid paralysis (AFP) cases and their healthy contacts in Pakistan in 2003. The clinical importance of CoSV remains unclear as data on epidemiology are scarce and no routine diagnostic testing is done. In this study, we characterized human CoSV (HCoSV) in a child with non-polio AFP and in sewage samples collected in Berlin, Germany. Using unbiased high-throughput sequencing and specific PCR, we characterized a HCoSV-D in stool samples of a three-year-old child hospitalized in Germany with non-polio AFP and travel history to Pakistan. The shedding pattern and absence of other relevant pathogens suggests that HCoSV-D may have been involved in the genesis of AFP. The HCoSV-RNA concentration was high, with 2.57 * 106 copies per mL fecal/suspension, decreasing in follow-up samples. To investigate the possibility of local circulation of HCoSV, we screened Berlin sewage samples collected between 2013 and 2018. Molecular testing of sewage samples has shown the presence of CoSV in several parts of the world, but until now not in Germany. Of our sewage samples, 54.3 % were positive for CoSV, with up to three viral species identified in samples. Phylogenetically, the German sequences clustered intermixed with sequences obtained globally. Together, these findings emphasize the need for further clinical, epidemiological, environmental, pathogenicity and phylogenetic studies of HCoSV. © 2021 Elsevier B.V.Veröffentlichung Not only biocidal products: Washing and cleaning agents and personal care(2018) Olsson, Oliver; Kümmerer, Klaus; Wieck, StefanieThe emission sources of biocidal active substances in households have been under discussion since these substances have been detected frequently in municipal wastewater and receiving surface water bodies. Therefore, the goal of this study was to investigate the products responsible for the emission of these substances to wastewater. We analysed the wastewater of two streets for a set of biocidal active substances. Time-proportional sampling was conducted for one week of each season during one year in each street. The 14 substances analysed with liquid chromatography coupled with tandem mass spectrometry were 1,2-benzisothiazol-3(2H)-one (BIT), C12-benzalkonium chloride, carbendazim, 5-chloro-2-methyl-2H-isothiazol-3-one (CMIT), dichlorooctylisothiazolinone (DCOIT), N,N-diethyl-meta-toluamide (DEET), diuron, icaridine, 2-octyl-2H-isothiazol-3-one (OIT), piperonyl butoxide (PBO), triclosan, tebuconazole, terbutryn and tetramethrin. Using data available from household product inventories of the two streets, we searched the lists of ingredients for the products possibly being responsible for the emissions. Except for four substances, all substances have been detected in at least 10% of the samples. Highest concentrations were measured for C12-benzalkonium chloride with an average concentration in the daily samples of 7.7ng/L in one of the streets. Next to C12-benzalkonium chloride, BIT, DEET and icaridine were detected in all samples in average concentrations above 1ng/L in at least one street. The results show that washing and cleaning agents were important sources for preservatives such as BIT and OIT, while triclosan was apparently mainly emitted through personal care products. The mosquito repelling substances DEET and icaridine were found throughout the year, with highest emissions in summer and autumn. In conclusion, the results demonstrate that the sources of biocidal active substances in municipal wastewater are complex and that measures for the prevention of the emission of biocidal active substances into the aquatic environment have to be carried out under different legislations. In einem von den Bundesländern und dem Bundesministerium für Umwelt, Naturschutz und nukleare Sicherheit finanzierten und koordiniert durchgeführten Untersuchungsprogramm wurden 49 Kläranlagen und ausgewählte Regenwasserbehandlungsanlagen auf prioritäre Stoffe und Biozid-Wirkstoffe untersucht. Ziel war die Schaffung einer validen Datenbasis zur Beurteilung der Relevanz urbaner Abwasserflüsse als Eintragspfade für Schadstoffe in die Gewässer. Die insgesamt 77 Stoffe waren unterschiedlich häufig im Ablauf der Kläranlagen und Regenwasserbehandlungsanlagen zu finden. Für 30 Stoffe konnten mittlere Ablaufkonzentrationen bestimmt und deutschlandweit anwendbare Emissionsfaktoren zur Quantifizierung der Stoffeinträge aus kommunalen Kläranlagen in die Gewässer abgeleitet werden.
